To improve the situation of cell attachment, we coated fibronectin on chitosan membranes. The cell morphology was observed by scanning electron microcopy (SEM). The MTT assays were made use of measurement of cell viability. Utilizing indirect immunofluorsecence analysis, cells were shown to express cytokeratin, rat parotid acinar cell specific secretory protein. Immunocytochemical staining of the immortalized cells involved the exposure to specific antibodies for the cytokeratins. Acinar cells have been difficult to maintain in primary or secondary cultures over extended periods of time. The rat parotid acinar cell adhesion increased with chitosan membranes coated fibronectin. In the next place, sialic acid added to the medium could promote parotid acinar cells activity increased. Salivary gland loss due to trauma, masticate betel nut or a variety of genetic disorders continues to affect most adults adversely at some time in their lives. The result of this study shows that this system would help us to generate an artificial salivary gland.
