In this research, I studied the effects of different degree of PEGylated adenovirus on its infection rate of target cells. I also compared the changes in physical properties and survival of PEGylated adenovirus under environmental stress. Adenovirus with different degree of PEGylation was used in the experiments. As the degree of PEGylation increased, the negative charge on the viral surface decreased and the more virus aggregation. Analyses of infection and attachment kinetics showed that the infection and attachment rates of virus on target cells decreased upon the increase in degree of PEGylation. It indicated that PEG on the surface of adenovirus possessed the shielding effect which resulted in decreasing the association of virus and the receptors on the cell membrane. Furthermore, studies in attachment kinetics suggested that the shielding effect of PEG on fibers of adenovirus may should be the most important factor in determining the infection rate.
