The most convenient means to determine the DNA concentration is to measure the absorbance at wavelength 260nm. In order to obtain a more precise measurement, we conduct an experiment, and based on a light-modulated spectrometer. The light was modulated at 1000Hz by a chopper and scanned into different wavelengths by the spectrometer. Under these conditions, we measure the absorbance at wavelength 260nm, and compare the results with those obtained by a GeneQuant pro RNA/DNA calculator. The two sets of results match each other. The absorbance standard deviation of GeneQuant pro RNA/DNA calculator is at wavelength 260nm, and the absorbance standard deviation of our light-modulated spectrometer is at wavelength 260nm. However, there are 2 possible errors in our system. One is the “Sudden Change” happened by the light of the Xenon lamp used. The other is the displacement error come from the instability of container.
