Description
Summary:博士 === 國立高雄師範大學 === 生物科學研究所 === 91 === Abstract Thyme(Thymus vulgaris) contains of essential oils favoring oxidation and anti-bacterial, especially thymol, is an antiseptic and disinfectant. As a matter of fact, most research has been concentrating on extracting the essence and its remedies. The following research adopted the thyme seeds to generate the aseptic plantlet---as the tissue culture. It is mainly to investigate the various concentrations of hormone supplied in forming the callus by stimulating hormone and gemma incubation in cultivating medium. The results demonstrate the experiment by using the calf thyme in forming the callus. With MS tissue culture , a BA 10 μM and NAA 0.5 μM accretion is the best among those. The greenish callus was incubated after two weeks. For the following two weeks observations, the gemma was formed and a whole plant was fully grown after a month. In tissue-transplanting experiment, cultivating the pure callus for two weeks, a colony of gemma has been propagated in BA 10 μM and NAA 0.5 μM. The colony covered almost 95% of the area. After finding out the optimum basic concentration of MS tissue culture, compared with the thyme leaves, nodes and internodes in BA 10 μM and NAA 0.5 μM, the results showed that the callus appeared in internodes the earliest. They were mostly white or yellow brownish in color. The overall speed of growing was slow. Propagation of plants through tissue culture has become an important technique to reproduce plants that are difficult to propagate conventionally by seed or vegetative means. However, a possible task will be fulfilled after the long run.
