It is an important virus infecting orchid plants and occurring worldwide. The purpose of this study was to sequence the genomic regions of ORSV Taiwan strain(ORSV-TW), and compare the nucleotide and amino acid sequences with those of four reported ORSV strains, ORSV-S1 (Singapore), ORSV-Cy1 (Japan), ORSV-Cy (Korea) and ORSV-U (America). As the result of sequencing, the full length nucleotides of ORSV-TW were 6,612 bp. The nucleotide sequences for 126/183 kDa RNA-dependent RNA polymerase(RdRp)gene were 3,336 and 4,839 bp which encoded proteins of MW of 126 and 183 kDa ,respectively,representing RdRp replicase and readthrough protein. Genes of movement protein (MP) and coat protein (CP) were 909 and 474, which encoded proteins of MW of 33 and 18 kDa, respectively. The nucleotide sequences of 5'' and 3'' non-translated region(NTR) were 63 and 414 bp, respectively. The results of comparison of nucleotide and amino acid sequences of ORSV-TW with those of reported ORSV-S1、ORSV-Cy1、ORSV-Cy、ORSV-U showed over 96﹪identity and similarity which proved that they are different strains of the same virus. In conclusion, the comparison of multiple alignments of nucleotide and amino acid sequences among ORSV strains displayed high identities in intraspecies. In the phylogenetic analysis,five strains of orsv had a closer relationship with. It is expected that the result of this study can help in the construction of the antivirus transgenic plants for controlling ORSV and detection of the virus in the future.
