When IBA was 10 mg/L, the length of root increased as the concentration of NAA increased. Medium with addition of charcoal could improve the growth of in vitro Phalaenopsis plantlets. Medium without charcoal appeared more vigous growth on the subterranean part. Nine different concentration of NAA and IBA combinations were used in Dtps. I-Hsin New Girl ‘KH 5250’ in vitro culture. The results showed that the number of roots increased rapidly at 8-10 week, then the rate slowed down at 10-12 week. Phalaenopsis plantlets were transferred to daughter-flask and cultured under 4 different light intensities. The plant size appeared significantly larger on plantlets exposed to 1000 Lux of light intensity. The hypogyny leaves of plantlets exposed to 3000 and 4000 Lux turned yellowish. The fresh weight, dry weight, root/leaf fresh weight ratio, root/leaf dry weight ratio, dry/fresh weight ratio, SPAD values, number of leaves, number of stomata, number of roots, diameter of roots, Fv/Fm of leaf and root appeared increasing as the light intensity increases. The fresh weight of Deflasked micropropagated plantlets of three Phalaenopsis hybrids were measured after stored at 20℃ for 3-12 days. The weight loss was 1.68-2.28% after 3 days, and 5.42-7.13% after 12 days. SPAD values and Fv/Fm showed no significant difference. The survival rate reached 96.4% after 10 weeks of growth. However, the plant size was smaller in Dtps. I-Hsin New Girl ‘KH 5250’ and Dtps. Leopard Prince ‘KH5733#2’ after stored for 9 and 12 days. Deflasked micropropagated plantlets of three Phalaenopsis hybrids were stored at 10, 15, 20, 25, 30℃ for 6 days. The weight loss was the highest (4.68-6.63%) when stored at 30℃. SPAD values and Fv/Fm showed no significant difference. SPAD values and Fv/Fm of Dtps. Leopard Prince ‘KH5733#2’ were low when stored at 10℃ and 30℃. After the storage, all three hybrids stored at 10℃ showed chilling injury. However, only Phal. Taisuco Kaladian ‘186’ stored at 15℃ showed chilling injury. The plantlets stored at 10℃ showed low survival rate (78.0-92.4%) for 10 weeks of growth and appeared slow growth due to chilling. The survival rate of other treated plantlets reached 90% or above and appeared normal growth. Different stuffing materials were used for the packaging box during storage. Only Dtps. I-Hsin New Girl ‘KH5250’ showed no significant difference in plant growth after 10 weeks of culture. The plant size of other two hybrids was smaller when absorbent paper was used.
