Summary: | 碩士 === 國立成功大學 === 生物科技研究所碩博士班 === 91 === Prostaglandins play an important role in regulating many physiologic reactions in mammals and exit in most tissues. In the previous studies, it is shown that prostaglandin F2α (PGF2α) can induce protein synthesis of neonatal rat cardiomyocytes and induce hypertrophy. Cardiac myocyte-derived cell line, H9c2, that maintains a cardiac-specific phenotype and undergoes hypertrophy in response to agonists at Gq-coupled receptors have not been clarified. The aim of this study was focused on whether PGF2a could induce H9c2 cells to undergo hypertrophy, and whether Anoectochilus formosanus HAYATA., a kind of Chinese herb, could cure or protect H9c2 cells from hypertrophy that induced by PGF2α. Results showed that protein concentrations of H9c2 cells after incubating with PGF2α (1μM) for 72 hr was 3 fold higher than control (without PGF2α). It also found that the effective doses of A. formosanus H. for the curing of cell hypertrophy were 1 mg/ml, 3 mg/ml and 4 mg/ml in crude extract, ethyl acetate extract and water extract respectively. Protecting effects of A. formosanus H. were found to be 4 mg/ml, 1 mg/ml and 4 mg/ml in crude extract, ethyl acetate extract and water extract respectively. In conclusion, PGF2α (1μM) can induce hypertrophy of H9c2 cells, and A. formosanus H. contains effective components which could cure and protect H9c2 cells from hypertrophy that induced by PGF2α.
