Bovine brain microvascular endothelial cells (BBMECs) were isolated by primary culture technique, and the cell suspension with density 3.0 × 104 cells/ml and cell viability higher than 92% was obtained. The BBMECs can be cultured to form a confluent monolayer, which serves as an in vitro blood-brain barrier model system retaining the characteristics in vivo. On the other hand, polybutylcyanoacrylate (PBCA) nanoparticles (NPs) and methylmethacrylate-sulfopropylmethacrylate (MMA-SPM) co- polymer NPs were synthesized, respectively, by emulsion polymerization and free radical polymerization. The investigation on the loading of stavudine (d4T), a human immunodeficiency antiretroviral agent, onto the two biodegradable polymeric NPs was performed. The experimental results indicate that the loading percentage (LP) of d4T onto MMA-SPM NPs was higher than that of d4T onto PBCA NPs under unadjusted pH (7.2). Freeze drying of the two NPs causes an increase in particle size and a decrease in LP of d4T. Under pH 7.4, LP of d4T on MMA-SPM NPs and PBCA NPs are, respectively, 51.65% and 58.66%. A variation in pH causes a decrease in LP for the two NPs. PBCA NPs are more stable for the loading of d4T than MMA-SPM NPs when variation of pH occurs.
