Summary:碩士 === 國立中正大學 === 化學工程研究所 === 91 === In this study, the effects of pH, the weight of magnetic nanoparticles, the ratio of carbodiimide/ADA, and the time of immobilization for adenosine deaminase (ADA) immobilized on magnetic nanoparticles is described. The optimal pH for ADA immobilized on magnetic nanoparticles is 5.8, the optimal carbodiimide/ADA ratio is 2, the optimal time is 30 minutes, and the optimal weight of magnetic nanoparticles is 110 mg. For this results, the percentage of ADA immobilized on magnetic nanoparticles is 99%. And then, the stabilities of immobilized ADA is also described. In this experiment, the immobilized ADA is more stable than free ADA, and it’s half life at 55℃ is 2.7 hours, one at 4℃ is 6.5 days. At last, the optimum conditions of the immobilized ADA catalyzing the deamination of adenosine is described. The optimum conditions are 55℃, 210 rpm, and pH 6.7. At these conditions, the kinetic parameter is obtained, and the Vmax (57 nmol/min), Km (0.36 mM) of Hanes plot is the most suitable value for this study.
