A series of batch test with different substrates (glucose, acetate, lactate, propionate) and concentrations (200, 300 mg/l as COD) was proceed to observe the substrate utilization rate. Combination with the microscope staining technology to confer the possible metabolism behavior. The results showed that under low phosphorus feeding, the phosphorus capacity of the sludge was lower compared with other phosphorus removal system under normal operation, only 1.82%. The results of the anaerobic batch test with different substrate showed that, the highest specific utilization rate was glucose(300),can reach 643.88 mgCOD/gMLSS*day. The lowest specific utilization rate was lactate(300) only 102.31 mgCOD/gMLSS*day。The specific utilization rate of glucose was higher than other acid (acetate, lactate, propionate). All anaerobic batch test of the sludge displayed phosphorus release and uptake behavior. The highest specific phosphorus release capacity in the anaerobic period was glucose(300), can reach 1.91 mg/g MLSS, the lowest was lactate(300), only 0.5 mgCOD/gMLSS. The results showed that even under low phosphorus feeding, The sludge of EBPR system still existence of PAO (Phosphate Accumulating Organism). About the variation of carbohydrate inside the sludge. Under high concentration of glucose feed, the carbohydrate inside the sludge will accumulate during the anaerobic period, there were no distinct change under low concentration. The carbohydrate inside the sludge will decrease during the anaerobic period with acetate, lactate, propionate feed. All batch test showed that the carbohydrate inside the sludge will increase during the aerobic period.
