However, there was less report about electro-ejaculation. For applying artificial insemination to endangered bird propagation, Tsaiya ducks were used as animal models to establish the method of electro-ejaculation. In the first experiment, our aim was to evaluate the effect on semen production whether the phallus was evaginated or not. If the phallus was evaginated under anesthesia prior to electrical stimulation, the semen volume per ejaculate was lesser than 0.02mL and the contamination rate was 57%. On the contrary, the average semen volume and average semen concentration was 0.13±0.07mL and 4.37±1.51×109/mL. The contamination rate was 33%. In the second experiment, two methods of semen collection from Tsaiya draks were compared: Manual massage (MM) and electro-ejaculation (EE). The average of semen volume per ejaculate and semen concentration were 0.14±0.11mL, 8.36±1.84(×109 /mL) in EE group, and 0.17±0.12mL, 6.17±1.83(×109 /mL) in MM group. The average of semen volume per ejaculate、semen concentration were not significantly different between EE and MM group (P >0.05). The proportion of fertile eggs in the second to fourth days and the second to eighth days following a single insemination were 85.40±12.71%, 67.93±18.51% in EE group and 68.71±14.46%, 44.01±14.20% in MM group. The average of duration of fertility in EE group (6.23±1.72 days) was longer than that in MM group (4.77±1.06 days). The average of duration of fertility and the proportion of fertile eggs were both no significantly different (P >0.05). In the final experiment, the method of electro- ejaculation was applied to captive mallards. The success rate of training ten drakes was 70%. The average semen volume and concentration per ejaculate was 0.13±0.06mL and 9±3.65(×109/mL). Ducks were inseminated with pooled semen (0.05mL, 1×108/mL). The fertile eggs appeared from the second to fifth days after a single insemination. The proportion of egg fertility and hatchability was 89% and 62.5% respectively.
