Summary: | 碩士 === 國立臺灣大學 === 園藝學研究所 === 90 === Citrus is one of the economical fruits in Taiwan. Citrus juice is also with largest production and market in the world. Liucheng, Ponkan and Tankan are the most important processing citrus cultivars in Taiwan, occupied approximately 74﹪of Taiwan’s citrus. In this study, trnL intron and trnL-trnF fragment in citrus were used for identification and classification, and later analyzed by UPGMA (Unweighted pair group method with arithmetic average), NJ (Neighbor-joining method), PA (Parsimony method ) and ML (Maximum likelihood method).
The results show that, Liucheng, Ponkan and Tankan are divided into 2 group and the molecular classification result are consistent with morphology classification. The variable fragment between oranges and mandarins in trnL-trnF were amplified and identified by DGGE (Denaturing Gradient Gel Electrophoresis). The method were capable of differentiating oranges and mandarins, but not for Haili Early and Tangelo. And for eight 100﹪ orange juices detection, seven were found to contain mandarin juice.
