Pollen viability is one of the main reasons. Successful inter-cross is closely related with the compatibility of the pollen grains on stigma, the capability of pollen grains growth in the style tracting tissues, and the successful entrance into the ovary for pollination. The pollen was observed by stain methods and in vitro or in vivo tests germination in order to found Oncidium pollination basic data for hybrid breeding. The results of this research are summarized as follow: it is clear that Oncidium pollen grains develop stop at tetrad stage and can not reach the true mature, so there is no starch accumulation. But the viability of the mitochondria and esterase had expression. When 3% sucrose were added to the culture medium, it was found that Oncidium pollen grains exhibited high viability. When using this culture medium to investigate different species pollen grains of the Oncidium, showed the different species pollen grains displayed different germination potential and viability. Angel ×Golden Shower "Liu Li " or Ramsey ×Golden Shower "Liu Li " belongs to the compatible inter-cross. After pollination, Golden Shower "Liu Li " pollen grains germinated about 2 days on stigma surface, grew into stigma about 5 days, grew into the upper ovary 8-11 days, and grew into the placenta 23-25 days.
