The mass production engineering of tissue culture became more important. The design and development for plant tissue culture vessel was important factor in the mass production engineering of tissue culture industry. The purpose of this study was to investigate the characteristic of several tissue culture vessels. According to the present standard equipments of tissue culture, ideal tissue culture vessel could be designed to supply for tissue culture industry in Taiwan. The investigated characteristics of tissue culture vessel included gas exchange rate, transmittance, spectral distribution of light and internal transmittance distribution. The result indicated that gas exchange rate about 0.025/hr could allow some gas exchange between outside air and inner air, and could isolate from water loss and contamination. The ideal material for tissue culture plastic vessel is polycarbonate. It has high light penetration, stable spectral distribution (transmittance about 0.9 in 430-760nm), autoclave-resistant, washable, and reusable. Cylinder vessel has the best light uniform condition of internal transmittance distribution of vessel. This study had investigated the requirements of the plant culture system included gas exchange rate, transmittance, spectral distribution of light, and internal transmittance distribution of vessel. Then the ideal design of tissue culture vessel also satisfy the required conditions of plantlet growth in vitro and fit to the standard specification of present tissue culture equipments.
