In order to elucidate the susceptibility as well as cause to the infection of Angiostrongylus cantonensis for different strains of mice, we compared the survival days of CBA、C57BL/6(B6) and BALB/c mice infected with 50 third stage larva of Angiostrongylus cantonensis per mice. The results showed that the B6 mice were survived after 30 days of infection, whereas the survival days of CBA and BALB/c mice were 25.3±0.58 and 28.0±2.31 days respectively. The worm recovery and length of worm in brain after 7、14 and 21 days of infection demonstrated that the worm recovery and the length of worm of B6 mice were the least among the three strains of mice (One-way ANOVA, p<0.05). The number of eosinophils in both CSF and blood all revealed that the B6 mice possessed the highest counts than the other two strains of mice (One-way ANOVA, p<0.01). We also measured both CSF and serum concentrations of specific antibodies (IgM、IgE、IgE、IgA、IgG3、IgG2b、IgG2a and IgG1) against the fifth stage larva of Angiostrongylus cantonensis by ELISA. The results showed that the serum IgM levels of BALB/c mice were higher than B6 mice after 7 and 14 days of infection. The serum IgG3 level of BALB/c was higher than CBA and B6 mice after 14 days of infection. The serum IgG2a concentration of BALB/c was significantly higher than B6 mice after 28 days of infection. The serum IgG2b level of BALB/c was higher than CBA mice after 7 days of infection. The CSF IgE concentrations of B6 were significantly higher than CBA and BALB/c mice (One-way ANOVA, p<0.001 and p<0.05, respectively). The results of immunoglobulin (IgG and IgE) antigen recognition revelaed that the CSF antibodies from early phase infection could react with 205 kDa of extracted fifth stage larva antigens. Furthermore, we also found that the CSF antibodies from late phase infection had antigen-binding reactivity with the lower molecule weight antigens. However, there was no specificity on antigen recognition sites among the three species of mice. These data suggested that BALB/c mice showed the greatest, CBA mice showed intermediate and B6 showed the lowest susceptibility to Angiostrongylus cantonensis infection. The dissimilar susceptibilty of the three strains mice to infection may be associated with the CSF IgE level produced by mice after infection. Yen CM 顏全敏 2002 學位論文 ; thesis 63 zh-TW |
