Osteoporosis is a serious problem for the postmenopausal women and aged populations. Several biochemical markers have been used for monitoring bone turnover. Among the markers of bone resorption, type I collagen crosslinked N-telopeptides (NTx) are derived specifically from bone collagen degradation and not metabolized. A ELISA (enzyme linked-immunosorbent assay)kit(osteomark®)was developed for measurement of the cross-linked N-telopeptides excretion. The monoclonal antibodies (mAb 1H11) used in the assay can recognize the epitope that is embodied in the N-telopeptide pyridinoline cross-linked domain of human type I collagen that is uniquely enriched in bone collagen. Surface plasmon resonance (SPR) technology has been applied for analysis of biomolecular interaction between antigen and antibodies. In this study, we immobilized the monoclonal antibodies mAb 1H11 on the surface of CM5 chip and detected their interaction with the urinary NTx excretion using the SPR-based method that we have established in our laboratory. Urinary samples of our subjects were assayed by both ELISA and SPR-based method. Results of both assays are in good correlation and are useful for further development of immunoassays to monitor the bone loss by detection of NTx in urine.
