The head and neck cancers, include tongue, larynx, pharynx, tonsils and nasopharyngeal carcinoma, are common diseases in Asia. In Taiwan, about 8% of cancer mortality, i.e. 2400 people each year, belongs to the head and neck cancer. In this study, colony and immunoflurescent staining assays were used to detect mutant frequency and molecular characterization of the HPRT gene in lymphocytes of patients with head and neck cancers before and after radiotherapy.
No significant difference was found for HPRT variant frequency (Vf) measurement between the colony assay and the immunofluorescence staining assay (P=0.754). A mean variant frequency of (3.37±2.14) ×10-5 of the HPRT gene in patients before radiotherapy. The variant frequency of HPRT gene is significantly increased in five patients (45%) during treatment, and in seven patients (78%) after treatment. The HPRT Vfs of patients, were elevated 3 folds to 13 folds, respectively, (p<0.01) at the end of radiotherapy. These data suggest that the HPRT variant frequency of head and neck cancer patients was increased about 6 folds after radiotherapy.
