Inhibition of angiogenesis, so called “anti-angiogenesis therapy” has emerged as a promising strategy for treating cancer since last decade. In these studies, we have examined the effects of Co 60 radiation and an antiangiogenic agent, TNP-470, on KHT sarcoma injected in the hind limb of C3H mice. Our morphologic evidences demonstrated that the death of tumor after radiation went through both dying mechanisms:necrosis and apoptosis. The degrees of necrosis and apoptosis caused by radiation were both dose- and time-dependent. Irradiation and TNP-470 decreased the vessel density of tumor in C3H mice. On day 35 after tumor cells injection, the average vessel number of one 200 X field vision in control mice was 38.4 ± 1.6. Whereas, 15 Gy radiation, TNP-470 alone and 15 Gy together with TNP-470 was significantly lower as 10.8 ± 0.8, 13.0 ± 4.0 and 8.2 ± 1.2 respectively (n = 2) (p < 0.05). Combined therapy using irradiation and TNP-470 prolong the lifetime of tumor-bearing C3H mice from 34.8 ± 6.1 days to 63.3 ± 3.8 days and 75.0 ± 11.7days (n = 4) (p < 0.05). Irradiation and TNP-470 also decreased the extent of lung metastases. On day 35 after tumor cells injection, the number of lung colony in control mice was 32.0 ± 16.9. Whereas, 15 Gy radiation, TNP-470 alone and 15 Gy together with TNP-470 was significantly lower as 12.3 ± 8.4, 9.8 ± 4.7 and 1.8 ± 1.7 respectively (n = 4) (p < 0.05).
