Hepatocytes have secretor functions that are polarized cell in liver organ. In our study, we use polarized hepatoblastoma cell HepG2 to investigate intercellular Calcium and cell proliferation after mechanical stress. Confluence and polarized monolayer cells were used physical touch to create wound on coverslip. In our results indicated that intercellular calcium wave propagated near the wound edge, and cells proliferation at later healing process. We used pharmacological reagents blocking intercellular calcium wave before wounding. We found that transient exposure of thapsigargin could completely inhibit the trauma-induced calcium wave. However, we found that intact liver also had intercellular calcium wave after mechanical stress. We try to use in vitro model asked why liver regeneration after hepatocytes were injure.
