Therefore the purpose of this experiment is to purify the enzyme a-D-galactosidases from rice. The formⅠa-D-galactosidases was purified by extraction , gel filtration on Sephadex G-75、G-100 column, and ion exchange chromatography on DEAE-Sephadex A-50 、CM-Sephadex C-50 column. Five a-D-galactosidases from rice were identified by ion exchange chromatography.It was purified with a 104-fold increase in specific activity. The yield was 24% and the specific activity was 1.04 unit/mg protein. The molecular weight of the enzyme was estimated to be 25 kDa by Sephadex G100 column.
