Summary: | 碩士 === 慈濟大學 === 毒理學研究所 === 89 === Trichloroethylene(TCE) and perchloroethylene (PCE), the important industrial solvents especially for dry clearing and degreasing, are environmental con- taminants. The pollution of TCE and PCE frequently occurs in workplaces, such as air and groundwater.It had been proved that TCE and PCE were harmful to the respiratory system since they are volatile organic solvents which are inhaled through airway. This study was to investigate the cytotoxic effects of TCE and PCE on alveolar macrophages obtained from piglet by bronchoalveolar lavage. TCE at the concentrarion of 1000 ppm significantly induced cell death. After alveolar macrophages treated with PCE for 6 or 12 hours, especially above 300 ppm, and 24 hours, especially above 100 ppm, PCEinduced increase in the release of lactate dehydrogenase (LDH) that was correlated to the decrease in cell viability, assessed by trypan blue exclusion. The result in this study indicated that TCE or PCE did not cause apoptosis on alveolar macrophages. Furthermore, PCE did not affect the release of prostaglandin E2 (PGE2) from macrophages when treated with lower concentration of PCE (0.01~10 ppm) for 24 hours. To summarize the injuries of TCE and PCE to the respiratory system , the cytotoxicity on the alveolar macrophages play an important role .
