In order to investigate the physiological roles of dietary fiber on intestine epithelium mucosal lymphoid tissue-peyer's patches (PP), 3-week-old male Wistar rats were fed with 8-week cellulose (5 and 10g/100g diet), rice bran (10 g/100 g diet), and wheat bran (10g/ 100g diet) as well as without any dietary fiber supplement as a control group. Plasma samples and secretory IgA of intestinal mucus were drawn for quantity of immunological proteins by enzyme-linked immunosorbant assay (ELISA).The distribution of B and T lymphocytes as well as IgA-plasma cells of PP observed by immunohistochemistry. When Wistar rats were fed with the diets containing various dietary fiber for 8 weeks, there was no significant difference among the five groups, neither in the food intake nor weight gain. Frozen section of immunohistochemistry had showed that there were more B lymphocyte staining in the germinal center of rats fed with dietary fiber compared to control group that B lymphocyte was dispersedly distributed in PP. There were also more T lymphocyte staining in the interfollicle area and dome area of PP in the rats with fiber treatment compared with control group. As B lymphocyte, there was IgA-plasma cell distributed circlely in the follicle of rats fed with dietary fiber but control group showed no pattern at all. Compared to the control group, the plasma IgG and IgA as well as sIgA detected by ELISA were significantly higher in the group fed with 5% cellulose. This results imply that dietary fiber not only stimulated differention of local immunity and lymphocyte migration, and also modulated the immune system in the whole system. Ying-Yueh Chu.
