Due to the potential toxicity of central nervous system, carcinogenicity and mutagenicity, the residue of nitroimidazole derivatives within the edible tissues need to be strictly controlled. The purpose of this study was to develop a high-performance liquid chromatographic (HPLC) method for the simultaneous determination of nitroimidazole derivatives within eggs and muscle of chicken. Tissue samples were ground and double extracted with acetonitrile. The supernatant of centrifuged samples were concentrated, and filtrated with a 0.45 µm filter membrane. The samples were cleaned up with n-hexane, a Sep-Pak® C18 cartridge, and eluted with methanol. The eluent was evaporated to dryness under a gentle stream of nitrogen at a temperature not exceeding 40℃. The residue was dissolved in 1 mL mobile phase for HPLC analysis. HPLC separation was carried out on a LiChrospher RP-18 column with a guard column and mobile phase consisting of methanol- KH2PO4 (10:90;v/v). The flow rate was 0.5 mL/min. The wavelength of UV detector was set at 320 nm. The average recoveries of spiking samples were 88.0-95.5% and 87.0-98.1% for chicken muscle and eggs, respectively. The detection limits of all drugs in all samples were 0.01 ppm. The average analytical time per sample was 110 minutes. Two hundred and eighty-eight samples were collected from traditional markets and supermarkets within three districts in Taipei area between April and May. The results showed that the residues of nitroimidazole derivatives (dimetridazole, metronidazole and ronidazole) were detected 13 cases in chicken meat and 17 cases in eggs.
