We cultured mitogen-activated immune cells, including Raw 264.7 macrophage cell line , primary peritoneal marophage and splenocyte with young rice grasses and survey the changes of immuno-indicator. In this study , effects of young rice grasses on inflammatory mediators and cytokines production, and cell proliferation of splenocytes were evaluated. First , the supernatants were harvested and tested for NO , PGE2 , IL-6 , IL-1b by the enzyme immunoassay. Our results demonstrated that three species of young rice grasses dose dependently reduced NO , PGE2 , IL-6 production in LPS and IFN-g co-activated Raw 264.7 culture. In primary peritoneal macrophages culture, NO and IL-6 production were also reduced, in accordance with Raw 264.7 cell line. Secondly, cytokines such as IL-2 , IFN-g , IL-4 and IL-5 from primary splenocytes were measured. The result showed that three species of young rice grasses decresded IL-4 , IL-5 secretion, however they enhanced IL-2 , IFN-g secretion in PHA activated splenocytes culture. Finally , the proliferative response of splenocytes was determined by [3H]-thymidine incorporation method. This result indicated that TCS 17 could promote proliferation of splenocytes. In summary, young rice grasses not only reduced inflammatory mediators and cytokines but also altered Th1 and Th2 cytokines secretions. It was found that TCS 17 had proliferative effect on primary splenocyte. Components of TCS 17 might contain a potent immunomodulatory substances that modulates the inflammatory responses , Th1/Th2 cytokines balance and splenocyte proliferation in vitro. The evaluation of immunomodulatory effects of Young rice grasses may provide an useful information for establishing a system for developing immunomodulatory component from food source.
