Summary: | 碩士 === 國立臺灣大學 === 昆蟲學研究所 === 89 === The mating behavior of Mythimna loreyi Duponchel occurs during scotophase. Production of sex pheromone in the virgin females follows a diel periodicity. Females start to synthesize sex pheromone in their first photophase, and the sex pheromone titer in pheromone gland on the scotophase was higher than on the photophase, and its titer was maximal on the third days after adult emergence. Females calling behavior is started with the onset of scotophase and this behavior was maintained till 1 hour into the photophase. Third-day old virgin females, at 10 hour into photophase, were used for pheromonal analysis such as Z7-12OAc, Z9-14OAc, and Z11-16OAc. Both subesophageal ganglion (SOG) extracts and synthesized pyrokinins/myotropins , which possessing the common C-terminal sequence FXPRL amide (X=V,T,S, or G), were tested for the pheromonotropic activity. The results show that maximal pheromonotropic response was exhibited with the treatment of 0.1 SOG equivalent. Dose-response results of the synthesized pyrokinins/myotropins show that Hez-PBAN, Lom-MT II, Lom-PK I, and Lom-PK II raised the sex pheromone titer significantly at the dose of 0.5~1 pmol/female. And Lom-MT I, LPK, Pss-PT, and type Iβ turn (cyclo) also raised the sex pheromone titer significantly at higher than 50 pmol/female doses. The functioning mechanism between peptide conformation and the pheromonotropic activity was also discussed.
