Summary:碩士 === 國立臺灣大學 === 化學研究所 === 89 === Glycosidases catalyze the hydrolysis of glycosidic linkages in aryl and alkyl glycosides, and occur widely in animals, fungi, plants, and bacteria. These enzymes’ crucial roles played in biological process involving cell to cell recognition and interaction, immune response, viral and bacterial infections, and so forth. Disorders of glycosidases may cause diabetes, cancer or AIDS, and so on. The design and synthesis of activity probes for glycosidases not only to label different enzymes, but can be treatment in clinical drugs.The concept is based on suicide inhibition of enzymes. A complete activity probe contains four moieties: (1) recognition head; (2) trapping device; (3) linker; (4) reporter group. We want to synthesize a series of activity probes about β-glucosidase, β-galactosidase, and sialidase. A series of activity probes carrying reporter groups of different properties, including dansyl group and biotinyl group.
