Antigens, porcine dermis homogenate (DH), pepsin treated type I collagen from porcine skin (PSCI-1), and non-pepsin treated type I collagen from porcine skin (PSCI-2), were injected into Balb/c mice and New Zealand White rabbits to give rise to an immune response. The characteristics of these antibodies were evaluated and the antigenecity of the porcine dermis and porcine dermal matrix was studied. From the results of the immune response of the mice, we found that PSCI-1 seemed to be more immunogenic than PSCI-2 and DH did. The solubility of the PSCI-1, PSCI-2 and DH may play an important role in such finding. The polyclonal antibodies,α-DH, α-PSCI-1, α-PSCI-2 and control antibody, were characterized by ELISA (enzyme linked immunosorbent assay) . The titer of α-PSCI-1 was higher than α-PSCI-2 and α-DH, the specificity of α-PSCI-2 and α-DH was higher than α-PSCI-1, and the sensitivity of each antibody was 1.2 ug/ml, 0.4 ug/ml, 1.2 ug/ml for α-PSCI-1, α-PSCI-2 and α-DH, respectively. The antibodies were used to detect the tissue section, The antigenecity of the porous dermal matrix was lower than untreated porcine dermis.
