Description
Summary:碩士 === 國立中興大學 === 園藝學系 === 89 === Summary It has been found in many researches that various forms of irradiation Could induce mutation in chrysanthemums, the rate of mutation can be substantially increased by the exposure of chrysanthemum plant material to γ-rays or X-rays. In practices,in order to speed up the production of new varieties and to enlarge the spectrum of colors, induction of mutation in chrysanthemum by radiation treatment has become a common practice in commercial breeder. The objectives of this experiment were to study the effect of γ-rays irradiation from cobalt 60 on chrysanthemum and selected mutant lines. Rooted cuttings of 13 cultivars and 9 selected color mutant lines of chrysanthemum(Dendranthema×grandiflorum(Ramat.)Kitamura) were treated with 10,15,20 and 25Gy of γ-rays,at a dose rate of 24.53Gy per minute. After treatment, the plants were pinched once and grown under protected construction. The radiosensitivity of all the irradiated plants were determined by the observation of morphological abnormalities of leaf, percentage of damaged growing point and percentage of survival, when flowering, data were recorded on the percentage of survival, frequency and spectrum of mutations. The radiosensitivity, with respect to different radiation damage and the mutation frequency, varied with the cultivars. The survival rate of chrysanthemum cultivars treated with 10,15,20 and 25Gy lay between 38﹪~100﹪,50﹪~100﹪,38~100﹪ and 0~100﹪ respectively, only one cultivar was lethal to 100﹪at 25Gy.The survival rate of selected mutants treated with 10,15,20and25Gy lay between 67﹪~100﹪,50﹪~100﹪and 0~100﹪respectively one selected mutant was lethal to 100﹪ at 25Gy. The mutantion frequency of cultivars at 10,15,20 and 25Gy were 20.2﹪,20.4﹪,21﹪ and 16﹪ respectively ,and for the selected mutants, the mutation rate at 10,15,20 and 25Gy treatment, were 11.8﹪,11.1﹪,11.3﹪and 4.5﹪ respectively. The mutation spetrum,i.e.the type of mutation induced, were greatly depends on the genetype of the treated material. and the range of flower color change induced by γ-rays also varietal depended. The purple color varieties shown the greatest range of flower color mutation after irradiation. The results showed that the production of new flower color and type of chrysanthemum can be achieved rapidly byγ-rays irradiation.
