Description
Summary:碩士 === 台北醫學院 === 醫學研究所 === 88 === Chitin, one of the most abundant polymers in Nature, is a biocompatible and biodegrable polymer. In the body, chitin can be digest through by enzyme and is a low immunogenetic poly-saccharide. Chitin can be separated in three types by its structure, with a-and b- types well known. To investigate a-chitin used, in this proposed research, we will apply the technique of wet spinning or casting to develop a facile type of chitin fiber and membrane from the chitin dope, which prepared with high molecular weight shrimp-resource chitin dissolved in metal salt / amine-containing solvent. At highly temperature and concentration, have low molecular weight .The stress is highly at 4% chitin solution. The chitosan scaffolds derived from chitin in the strong base and high temperature bs used for tissue engineering use. In the X-ray diffraction pattern of chitin, some differences were observed between a- and b- types. In the MTT assay, we can find chitin is biocompatible and bioabsorbable. In the thermal analysis, rH of carboxymethylchitin found at temperature 217°C different from other types. We also find the characteric absorbant peak by FT-IR . After enzyme immobolization of chitin membranes, found the residue activity about 0.2-0.3 units at 1 cm2 membrane by the spectrumeter. We hope that the uses of carboxymethylchitin for enzyme immoblization, in the future, can have a good application at the biosensor.
