The design and synthesis of an activity probe of PTPases are described here. The concept is based on suicide inhibition of enzymes. This mechanism-based activity probe adopts a cassette-like design with a phosphate group serving as the recognition head. A complete activity probe contains four moieties: (1) recognition head; (2) trapping device; (3) linker (4) reporter group.
