The design and synthesis of an activity probe of PTPases are described here. The concept is based on suicide inhibition of enzymes. This mechanism-based activity probe adopts a cassette-like design with a phosphate group serving as the recognition head. A complete activity probe contains four moieties: (1) recognition head; (2) trapping device; (3) linker (4) reporter group. A series of activity probes carrying reporter groups of different properties, including diethyl- aminocoumarin derivative, dansyl group and biotinyl group, were prepared in this thesis Lo, Lee-chiang 羅禮強 2000 學位論文 ; thesis 110 en_US
