Summary:碩士 === 國立臺灣大學 === 化學研究所 === 88 === Protein tyrosine phosphatases (PTPases) widely exist in almost all sorts of creatures and are an important class of enzymes in regulating various biological processes. The design and synthesis of an activity probe of PTPases are described here. The concept is based on suicide inhibition of enzymes. This mechanism-based activity probe adopts a cassette-like design with a phosphate group serving as the recognition head. A complete activity probe contains four moieties: (1) recognition head; (2) trapping device; (3) linker (4) reporter group. A series of activity probes carrying reporter groups of different properties, including diethyl- aminocoumarin derivative, dansyl group and biotinyl group, were prepared in this thesis
