The characteristics of the 99mTc-triamine phenol complexes obtained was investigated by solvent extraction, TLC, paper electrophoresis and HPLC.
The lipophilicities of the 99mTc complexes are generally poor but being significant for those with diethylenetriamine backbone. The labeling efficiencies can reach more than 80% in physiological and basic pH media for most of the 99mTc complexed preparations. The 99mTc complexes with N-bounded pyridinyl seem to show more stable than those without N-bounded pyridinyl. One of the 99mTc complexes with N-bounded pyridinyl demonstrates to have prominent positively charged species as high as up to 60% - 80%. The cationic species formed are hydrophilic and partition is prominently favored to the aqueous phase in the CH2Cl2/buffer extraction system.
Biodistribution of four technetium-99m labeled complexes were performed with the C3H/HeN mice. Considerable uptake of the four 99mTc complexes by myocardium was observed but the myocardial/blood ratios detected being generally lower than 1.
