The 157 kinds of herbs, collected from a herbal garden which belongs to our department were tested on ellagitannin components using thin layer chromatography (TLC) method. There were 48 herb¢s leaves showing positive ellagitannin coloration, and the lyophilized powders of their aqueous portion of 70﹪acetone extract solutions were chosen for evaluation of antihepatotoxic activities by using in vitro rat primary cultural liver cell model. The twenty of 48 herbs exhibited well antihepatotoxic activity against liver injury induced by carbon tetrachloride. However, the in vitro antihepatotoxic activity was reconfirmed with direct transaminase inhibition test. Five herbs, including Macaranga tanarius (Linn.) Muell.-Arg., Terminalia catappa Linn., Geranium nepalense Sweet var. thunbergii (Sieb. & Zucc.) Kudo, Ludwigia octovalvis (Jacq.) Raven. and Securinea virosa (Roxb.) Pax et Hoffm., had indicated real hepatoprotective activities, which were considered to have both a strong in vitro antihepatotoxic activity and a lower direct transaminase inhibition. Furthermore, the hepatoprotective effect of 5 herbs was studied on in vivo animal model of ICR mice, which liver injury were also induced by carbon tetrachloride. The leaves of Macaranga tanarius (Linn.) Muell.-Arg. have illustrated conclusively the best hepatoprotective activity among 5 herbs. Therefore, the aqueous portion of 70﹪acetone extract solution of this herbal leaves was extracted successively with ether, ethyl acetate and n-butanol, and remained a water layer. The n-butanol layer showed the best in vivo antihepatotoxic activity in ICR mice compaired with those of other 3 layers. Ellagitannin components were isolated from the n-butanol layer by using column chromatography and high performance liquid chromatography. A pure compound which retention time is 16.5 min in RP-HPLC analysis spectral profile was obtained and displayed an excellent antihepatotoxic activity on ICR-mice of carbon tetrachloride-induced liver injury.
