Tissue factor(TF), a special mediator expressed on monocyte, initiates extrinsic coagulation pathway and may play an important role in the process of DIC. Andrographolide, an extract from Chinese herb andrographis paniculata, has inhibitory effect on PAF-induced platelet aggregation. Moreover, it has been suggested that TF expression may be significantly induced by PAF. Thus, The aim of this study is to investigate the effects and the possible mechanisms of andrographolide on LPS-induced tissue factor expression in human monocytic THP-1 cell. Results demonstrated that pretreatment with andrographolide(20~100 mM) inhibited the LPS (1 mg/mL) induced TF expression and activity in a concentration-dependent manner in THP-1 cell. In addition, , andrographolide(200 mM) did not affect cell viability. Furthermore, andrographolide significantly decreased the LPS-induced NO production. Besides, andrographolide attenuated the LPS-induced IkBa degradation in a dose-dependent manner. In conclusion, andrographolide suppresses LPS-induced TF expression and activity in human monocytic THP-1 cell.
