We used the nontoxic Pseudomonus Exotoxin (PE) as a carrier protein to enhance the immunogencity of GnRH by chemical conjugation. In trial 1 the male chicks were divided into four treatments:Control, intact untreated; Castrate, intact castrated untreated; GnRH, intact immunized against GnRH; PBS, intact immunized control. The immunization schedule was carried out at 3、8、16 weeks of age. Sera from roosters were reactive in enzyme-linked immunosorbent assays (ELISA). Unfortunately, anti-GnRH antibody titer didn’t significantly develop (p>0.05) after immmunization. In trial 2 the male chicks were further divided into eight treatments:Control, intact untreated; Castrate, intact castrated untreated; OG、CG, intact immunized with mGnRH-PE in egg (OM) or male chickens(CM); OC、CC, intact immunized with cGnRH I-PE in egg (OC) or male chickens (CC); OO、CO, intact immunized with PBS in egg(OO) or male chickens (CO). The immunization schedule of egg (OG、OC、OO) was 18 day of incubation、3、15 weeks of age.That in chickens (CG、CC、CO) was 1、4、15 weeks of age.The results showed that the PE was an effective immunogen to induce high antibody in roosters. Meanwhite the GnRH obtained from mammals could also induce sustantial antibodies in poulty. Although GnRH-PE could induce antibodies, it seemed that the titer was not sufficient to inhibit the gonadal development in roosters. To develope an effective immunocastration vaccine for roosters, it needs further studies on optimizing the immunization protocols.
