Summary:碩士 === 中原大學 === 醫學工程學系 === 88 === The changes of surface glycoprotein Ib expressions of platelets were applied to index the activation of platelets for evaluating the blood compatibility of different biomaterials。With flow cytometric method,we measure the difference of MFI(mean fluorescence intensity) for two categories of biomaterials:a) PU(polyurethane),PU-PEG(polyethylene glycol) and PU-PEG-Heparin;b) ePTFE(expanded polytetrafluorethylene) and modified ePTFE with C2H2/N2 by 30W and 1 minute of plasma treated under static and shear flow conditions。 The results showed that ΔMFI were 174.777.8,61.338.5 and 75.134.4 for PU,PU-PEG and PU-PEG-Heparin,respectively。The modification of PU surface was significantly reducing ΔMFI values (p<0.05,n=5)。Whereas,under flow conditions,ΔMFI for PU and its modified materials were not significantly difference although that for PU was still the highest among those materials。ΔMFI for plasma treated ePTFE were significantly higher(p<0.05,n=4) than those for non-treated ePTFE for both A and C types。For example,ΔMFI were 131.853.2 and 138.361.7 for non-treated A and C type of ePTFE,respectively,whileΔMFI were 170.742.3 and 185.962.2 for plasma treated cases。 In conclusion,ΔMFI of surface glycoprotein Ib of platelets could detect the activation of platelets stimulated by the surface of biomaterials under static and flow conditions。Further exploring the technique for blood compatibility test may be valuable。
