It may be one of the elements in the regulation of nucleolar function. Recent studies have shown that protein B23 forms a specific complex with the human immunodeficiency virus type I (HIV-1) Rev protein. Furthermore, it is reported that RevNLS peptide (ARRNRRRRWREYC), a synthetic peptide containing the NLS sequence of HIV-1 Rev protein, has a high affinity to protein B23. Hence, it is the first time to examine the biological effects of RevNLS peptide on human promyelocytic leukemia HL-60 cells. Sodium butyrate (BuONa) is a potent differentiation inducer and cell growth inhibitor. And BuONa had been clinically applied on treating human leukemia in past years. Recent studies have shown that BuONa could also induce apoptosis. In this study, attempts are made to determine whether or not the biological effects of RevNLS peptide be enhanced after BuONa pretreatment. Our results suggest that (a) high dose of RevNLS peptide has a lethal effect on HL-60 cells, but low dose of RevNLS peptide has little effect on HL-60 cells (b) low dose of RevNLS peptide has an enhanced effect on apoptosis after BuONa pretreatment and nucleophosmin/B23 protein levels are dramatically decreased during RevNLS peptide/BuONa-induced apoptosis. 翁一鳴 2000 學位論文 ; thesis 50 en_US
