Summary: | 碩士 === 東吳大學 === 化學系 === 87 === Microdialysis sampling has become an important method for the continuous monitoring from an in vivo environment. This technique has been used to monitor many exogenous species such as drug substances and thus be widely used in pharmacokinetic study.
HPLC/electrospray/mass spectrometry (HPLC/ES/MS) has emerged as an effective and useful analytical technique for many applications. In the first part of this study, we developed an HPLC/ES/MS method for the analysis of cocaine and it’s metabolite benzoylecgonine in rat’s brain with microdialysis. The sensitivity, linearity, detection limit, sample stability, method precision and accuracy associated with this method are reported and pharmacokinetic profiles of cocaine in rat’s brain are also described.
Secondly, We developed an HPLC/ES/MS method to study the pharmacokinetics of flunarizine in the whole brain of Sprague-Dawley rat. In addition, we adapted this method for microdialysis application. The linearity and method precision and accuracy were investigated and the method was applied to pharmacokinetic study of flunarizine in rat’s brain with microdialysis.
