Characterization of a Sorghum cDNA Encoding Soluble Starch Synthase in Endosperm
碩士 === 國立臺灣大學 === 農藝學研究所 === 87 === A partial cDNA of 2230bp (accession no. AF168786), encoding a soluble starch synthase (SSS), has been cloned with 3'' RACE, 5'' RACE and RT-PCR techniques from sorghum (Sorghum bicolor (L.) Moench) seeds. This cDNA contained a 1569bp open readi...

This cDNA contained a 1569bp open reading frame with 523 amino acid residues. This partial putative amino acid sequence didn''t include the transit peptide. The deduced amino acid sequence of SSS shared 97.1% and 87.2% identity with maize starch synthase I (SSI) and rice SSS, respectively. The putative SSS amino acid sequence contained a highly conserved KXGGLGDV region, a putative ADP-glucose binding site. There were introns present in sorghum SSS gene, as revealed by the results of genomic PCR. The arrangment of these introns in sorghum SSS gene may be similar to that in rice. The sorghum SSS gene mainly expressed in seeds and leaves. The expression of SSS gene began at 5 days after flowering (DAF) in seeds and reached the highest level at 15 DAF. Southern blot analysis suggested that SSS was encoded by a single copy of gene. Hsieh, Jaw-Shu 謝兆樞 1999 學位論文 ; thesis 77 zh-TW |
