This cDNA contained a 1569bp open reading frame with 523 amino acid residues. This partial putative amino acid sequence didn''t include the transit peptide. The deduced amino acid sequence of SSS shared 97.1% and 87.2% identity with maize starch synthase I (SSI) and rice SSS, respectively. The putative SSS amino acid sequence contained a highly conserved KXGGLGDV region, a putative ADP-glucose binding site. There were introns present in sorghum SSS gene, as revealed by the results of genomic PCR. The arrangment of these introns in sorghum SSS gene may be similar to that in rice. The sorghum SSS gene mainly expressed in seeds and leaves. The expression of SSS gene began at 5 days after flowering (DAF) in seeds and reached the highest level at 15 DAF.
