The mini-scales of in vitro stock plant were sliced into 5mm as explants inoculated on MS medium supplemented with auxin and cytokinin. It was demonstrated that sole NAA 0.1~0.5 mg/L is effectively to induce direct caulogenesis, and regenerated 5.0 bulblets per explant. The addition of BA 0.05~0.5mg/L showed slight stimulation of adventitious bud formation, achieved 1.2 bublet, but pronouncely inhibited rhizogenesis. The combination of BA 0.05~0.5mg/L with NAA 0.1~0.5mg/L showed negatively antigonistic to auxin effect, reduced bud number and scale leaf emergence of the bulblets. In evaluation of propagation efficiency and regenerated bulblet growth, the combination of NAA 0.1mg/L and BA 0.01mg/L is suitable for two tested lily hybrids. The anatomical study showed that the position of regeneration is limited to the adaxial surface, the peripbery cell degraded the storage amyloplasts, and destined to caulogenesis forming adventitious bud primordium about 10 days after culture, while leaf primordial differentiation was also initiated rhizogenesis in the proximal end. The induction of indirect regeneration were also conducted in these two LA and LO hybrids. Both picloram and dicamba 3mg/L were capable to stimulate more than 90% callogenesis in mini-scale slices. The picloram was more efficient to proliferate yellow friable calli and numerously globular embryoids. The friable callus was transferred into 1/2MS liquid medium containing 2,4-D 2mg/L and BA 0.5mg/L, obtained plantlets in less than one months. The LA and LO scale explants were presoaked with colchicine 0.05~0.2% or oryzalin, then inoculated on regeneration medium. The root tips of bulblet were collected for cytological examination. 4 plantlets of colchicine 0.1% pretreatment were assessed as tetraploid 2n=48, the stomata size of scale leaf was measured as 150.9μm larger than the 110.52μm of diploid plantlets.
