Most patient with osteoarthritis are the elder, who are also at the risk of osteoporosis. Osteoporosis is the net bone loss from that bone formation from osteoblast lower than that of bone resorption from osteoclast. The NSAIDs used in the treatment of osteoarthritis may damage the articular artilage, and whether have any influence on biological activity in osteoblast is still unknown clearly. Recently we found that cyclooxygenase have two isoforms: COX-1 and COX-2. Indomethacin is the selective COX-1 inhibitor and meloxicam is the selective COX-2 inhibitor. The purpose of this study is to understand different NSAIDs whether have any influence on cell growth and biological activity in human osteoblast. We take spongy bone from patient with knee osteoarthritis undergoing TKA to harvest osteoblast for culture. Using the primary human osteoblast model culture, we compare the effects of different concentration of indomethacin and meloxicam on cell growth and biological activity in osteoblast with MTT analysis and osteocalcin concentration. The results show that (1.) indomethacin and meloxicam have trend to increase cell growth at the concentration of 5x10-6M; (2.) indomethacin and meloxicam have no influence on osteocalcin concentration in osteoblast at the concentration of 10-6M and 5x10-6M within 10 days culture; (3.) indomethacin and meloxicam have trend to inhibit endogenous prostaglandin E2 secretion from osteoblast.
