Recently, it was reported the effects of dietary ellagic acid on rat hepatic and esophageal mucosus cytochrome p450 and phaseⅡenzyme. There is no any aviable informations to address the effect of ellgic acid on cell cycle and N-acetyltransferase activity in human bladder tumor cells. Ellagic acid was used to determine the inhibition of cell growth (T-24 cell line) by trypan blue exclusion and revealed ellagic acid inhibited the human bladder tumor cell proliferation. Ellagic acid also induced DNA damage and morphology changes in this examined tumor cells. The DNA content analysis in human bladder tumor cells was determined by flow cytometry. The results also show that the treatment of ellagic acid in the cell culture results in the inhibition of S phase in cell cycle. The proportion of cells in S phase progressive decreased concomitant with an increasing G1 phase cells. After 12 hr incubation, ellagic acid treatment at 0.5-10 mM of bladder tumor cells (T-24) obtained the obvious inhibition in S phase. The degradation of genomic DNA was also analyzed in response to ellagic acid treatment by size fraction migration in T-24 cells. Ellagic acid could degraded the DNA results in the DNA damage.
