Summary: | 碩士 === 大同工學院 === 化學工程研究所 === 86 === Polyvinyl alcohol (PVA) has advantages of none-toxic and low cost in comparisonwith other immobilized materials. As shown in the results of this study, whenthe cells of Gluconobacter oxydans and the mycelium of Aspergillus japonicuswere immobilized with 10% (W/V) PVA, 51.72% and 55.84% of enzyme activity 0.6and 0.65 of effective factor for B-fructuranosidase and glucose dehydrogenasewere expressed respectively in the PVA gel. However, mass transfer of oxygen molecules in immobilized PVA particles was so low as to decrease the rate ofreaction.Breaking the immobilized particles could effectively raise the apparent activityup to 72.63% and 85.41% respectively, and make the reaction rate of whole systemup to 60% of which when reacted with free mycelia and cells.Inaddition, a complex cells bioreactor was employed, with microfiltration systemto produce high content fructooligosaccharides continuously. In this bioreactor, mycelia of Aureobasidium pullulans, which has B-fructofuranosidase and livingcells of Gluconobacter oxydans, which has glucose dehydrogenase, were reacted together with sucrose solution. This high content fructooligosaccharide production system maintained consistent temperature, aeration and agitation, continuously filtering the products through microfilter out of the bioreactor, and synchronously adding sucrose solution into bioreactor. Such reaction systemsould continuously produce high content fructooligosaccharides up to 80% in dryweight basis over 7days.
