The chronic type Bhepatitis may evolve a healthy carrier, cirrhosis or hepatocellular carcinoma.The sequence of HBeAg from sera of patients with hepatitis and thesequence of the entire core antigen of liver-derived HBVs in hepatomapatients reveal the high frequency mutations at the highly evolutionarilyconserved regions. There may had a relationship between the regions ofantigenic determinant of HBeAg and the high frequency mutation spots ofthe liver-derived and nature-occured HBV. To study this correlation, theform of antigenic epitope of HBeAg was investigated. This study mayexplain that HBeAg variants may accomplish immunoevasion via escapemutation along the antigenic determinants of HBeAg. The antigenicdeterminant of the HBeAg was mapped using monoclonal antibody. Sevenpotential epitopes of HBeAg were analyzed by hydrophilicity andsynthesized as peptides. Recombinant protein, SV, was tested for specificantigenicity with monoclonal antibody and sera from patients withhepatoma and used as a target protein for the binding with monoclonalantibody. The blocking activity of each peptide in the binding ofmonoclonal antibody to the target protein was measured by Western blotimmunodetection and ELISA. No blocking effect of peptide was identifiedin both assays.
