The results obtained were summarized as follows: 1. The emergence of buds, which were incubated on MS medium for 3 weeks, were not significantly different among the treatments of different concentration of agar, and the treatments of activated carbon, plant regulator, and non-plant regulator, in the basal medium. 2. The length and diameter of shoot in Anoectochilus formosanus seedlings incubated in low concentration of agar was better than in high concentration ones. Under medium containing without plant regulator and activated carbon, while leaf growth of seedling was increased greatly in medium with high concentration of agar.The diameter of shoot in seedling was promoted with medium containing BA and NAA, while the length of shoot was reduced except with medium containing 4g/l agar. 3. The length of shoot was reduced greatly with medium containing 3% of activated carbon and 4g/l of agar, but the diameter of shoot was promoted mostly. The leaf growth of this seedlings was enhanced with medium containing 12g/l of agar. The diameter of shoot was increased by using medium without adding activated carbon. 4. The resul t indicated that the medium containing 3mg/l BA, 0.2mg/l NAA and 4g/l agar was the most avaiable medium to greatly promote the length and diameter of shoots in seedling incubated from stem segment of Anoectochilus formosanus. On other hands, the leaf growth was promoted with medium containing 12g/l agar without plant regulator.5. The stem segment of Anoectochilus formosanus was incubate d on medium containing the three level of PEG for two months. The length and d iameter of shoot was reduced with medium contain 15% and 25% of PEG, when it c ompared with the non-PEG treatment. The length and diameter of shoot in seedli ng, which was incubated on medium with 15 and 25% of PEG, was not promoted wit h the application of 3mg/l BA and 0.2mg/l NAA.6. When the seedlings were incu bated on MS medium and Hyponex No.1 without the application of activated carbo n:the number of adventitious bud in seedlings was induced with medium contain ing 3mg/l BA, while this was not obtained with medium containing 0.2mg/l NAA. The number of adventitious bud and fresh weight of seedling was increased with medium containing 3g/l tryptone. The growth of seedling was promoted with the application of 3g/l tryptone and 2g/l peptone adding to the basal medium. The medium with tryptone have enhanced the increasing of fresh weight of seedling s more than the medium containing peptone. The adventitious bud of seedling wa s not promoted with medium containing activated carbon in MS and Hyponex No.1 medium, but plant height and the leaf growth of seedlings was greatly enhanced .
