Description
Summary:碩士 === 國立臺灣大學 === 微生物學研究所 === 86 === To identify the putative specific antigens from Helicobacter pylori that isiso ated from gastric specimens of the mucosa-associated lymphoid tissue lymphoma MALToma), secreted proteins of H. pylori were compared on SDS-PAGE by Westernb lot using sera from patients with MALToma. A 19kDa. protein was seen in strain sisolated from patients with MALToma but not in strains from those with other diseases. The protein was purified and amino acid sequences were determined by microsequencing.Sequences comparision revealed it was a FldA homologue. DNA se quencing in sixteen bacterial strains revealed that insertion of a nucleotide "G" at position 481 ofthe fldA gene were more frequently observed in strains associated with MALToma than those with other diseases (8/8 vs 2/8, p<0.01). T he fldA gene from a strain associated with MALToma was cloned and recombinant protein was expressed in E. coli.Serum samples of 12 out of 17 (70.6%) patient s with MALToma were positive for theantibody to the recombinant FldA protein w hile 7 out of 42 (16.7%) patients with other diseases were positive (12/17 vs 7/42, p<0.0001). These results suggest that this antigen of H.pylori may be as sociated with gastric MALToma. It may playa role in the pathogenesis of gastri c MALToma. Antibody to this antigen may be a serologic marker of the disease.
