In germinating maize kernels, however, beta-amylase is expresse d in the aleurone cells, which contain no starch for its exo-amylolytic action . Difference in the localization of enzyme and substrate molecules makes it d ifficult to understand the physiologicalfunctions of beta-amylasein vivo. To further investigate its functions, maize genomic libraries were constructed, a nd a full-length beta-amylase gene was cloned and sequenced. The primary stru cture of beta-amylase was also deduced from the sequence data. Beta-amylase p romoteractivity analysis was also undertaken with transient expression systems . Three different systems were tested, but proved unsuccessful. The promoter sequences were also analyzed by searching through internet resources and rese arch articles. Possible cis-elements in promoter region were found by databas e searching. Moderate degrees of homology were also found by computer-aided a lignment around TATA box and 5''-UTR region among beta-amylase promoters of mai ze, barley, and rice.
