There are differ distribution of many ethnic groups in the world In both polymorphisms. They were also associated with medicine metabolism and susceptibility to neoplasm, for instance lung cancer. In the ethnic distribution of genotype study, subjects are junior high school students of aborigines contained Paunch (n=169), Puyumar (n=38), Bunun (n=72), Atayal(n=72), Paiwan(n=48), min-nan(n=57) and Philippines(n=120). In lung cancer susceptibility and genotype study, subjects are woman lung cancer patients without smoking(n=31) and control(n=61). Genomic DNA were extracted from blood samples from subjects. The target DNA fragment: were amplified by PCR, and genotype were determined by RFLP. The subjects of previous two study were interviewed by structure questionnaire, to understand another environmental factors. The result of two study, the genotype distribution of aborigines, case and control were conformed by Hardy-Weinberg equilibrium. In Val polymorphism, the proportion of variant genotype (Val/Val) is Philippines 2.52%, Paunch 3.3%, Paiwan 4.9%, Atayal 6.3%, Bunun 8.1%, Puyumar 11.4%, min-nan 16.2%. There are difference of Val polymorphism among Philippines, Paunch, Paiwan and min-nan. In MspI polymorphism, the proportion of variant genotype (m2/m2) is Puyumar 8.8%, Paiwan 11.1%, Philippines 11.3%, Paunch 12.6%, Atayal 16.4%, Bunun 21.7%, min-nan 24.5%. There are difference of MspI polymorphism among Puyumar, Philippines, Paunch and min-nan. In another study, the odds ratio of m2/m2 and m1/m2 versus m1/m1 genotype for lung cancer is 1.05 (0.37-2.95), and odds ratio which adjusted by demographic variable is 0.91 (0.24-3.49 ). The odds ratio of Val/Val and Ile/Val versus Ile/Ile genotype for lung cancer is 1.06 (0.37-3.5) , and odds ratio which adjusted by demographic variable is 1.41(0.40-4.95). The result doesn''t show statistic significant. Adenocarcinoma of case who categorized by histological type still doesn''t show statistic significant between genotype and Adenocarcinoma. The limitation of study is less samples. Furthermore, the future of study could research mutiple-gene and individual susceptible for lung cancer, and interaction between environment an gene. Key words: CYP1A1 polymorphism , aborigines , lung cancer Ying-Ching Ko 葛應欽 學位論文 ; thesis 67 zh-TW |
