Summary:碩士 === 淡江大學 === 化學學系 === 85 === A mixed-valence cluster of chromium hexacyanoferrate based biosensor .for the determination of glucose was demonstrated. The cluster was prepared by scanning potential of a rotating glassy carbon electrode between -0.2V~ +0.95V in 0.05M tetrabutylammonium p-toluenesulfonate solution containing 20mM Cr(NC)3)39H20 and 5mM KsFe(CN)6 for 30 minutes. The glucose oxidase was added the aforementioned solution and codeposited on the glassy carbon electrode with the same condition for 10 minutes. Subsequently, the glucose can be measured at 0.0V(vs. Ag/AgCl), in 0.1M KCI, 0.05M succinic, pH6 buffer solution, 400rpm, 25 ℃, the linearity of the response is up to 4mM(with correlation coefficient 0.9994). The sensitivity of the electrode is 3.793μA/mM-cm2. The estimated detection limit (S/N=3) is 5x10(-6)M. The response time 195% is 33 seconds. The standard deviation of twenty repetitive detection of 5x10(-4)M glucose is 2.11%. There is no significant interference observed from the addition of 0.2mM easily oxidized compound, such as uric acid, acetaminophen, tyrosine, galactose at the presence of 5x10(-4)M glucose while, a limited interference were abserved from both ascorbic acid and cysteine at 1.48% and 2.78%, respectively.
