Highperformance liquid chromatography (HPLC), gas chromatography (GC)or gas chromatography/mass spectrometry (GC/MS) has been widely used for the determination of amphetamine and related compounds in biological fluids .The purpose of this study is to evaluate the effectiveness of HPLC/ES/MS for the analysis of methylamphetamine, amphetamine, ephedrine, and phenylpropanolamine in human urine sample. HPLC/electrospray/ MS (HPLC/ES/MS) has emerged as an effective and useful analytical technique. ES ionization produces ions at atmospheric pressure and relatively low temperature which minimizes thermal decomposition of labile compound. Furthermore, aqueous sample can be analyzed with little or no sample preparation. In this study, we developed a LC/ES/MS method for the detection of amphetamine and related compounds. The effect of ES/MS capillary exit (CapEx) volts, solvent composition, and modifier on the detection were investigated. A simple but effective sample preparation technique was developed for urine sample. It utilized microfiltration membrane to remove proteins and other compounds in urine prior to LC/ES/MS analysis. Good linear range (100~1000 ng/ml) and low detection limit (10 ng/ml, S/N=3) were achieve by SIM. Finally, this method will be applied to analyze the urine samples from drug addicts.
